PhosphoBiomarkers in Oncology
Phosphorylated, or activated, forms of proteins play an important role in the growth and progression of cancer. TMD has developed specialized assays for detecting the activated forms of PhosphoBiomarkers in preclinical and clinical samples.
Antibodies for the Detection of PhosphoBiomarkers
TMD can provide consultation for selecting the best in class commercial antibodies for the detection of PhosphoBiomarkers. Antibodies are evaluated based on specificity for a given phosphorylated antigen, and tested in cell line controls as a part of assay development and validation.
PhosphoGuard™
TMD’s PhosphoGuard™ Fixative has been used to collect formalin fixed biopsies in many clinical trials worldwide, including numerous trials involving Tykerb.
Our customers have always liked how easy the PhosphoGuard™ kit is to use, however the kit needed to be shipped on dry ice and stored frozen. Through extensive stability testing, the PhosphoGuard™ kit can now be shipped and stored at room temperature, thus freeing up valuable freezer space in the laboratory.
Phosphorylation and de-phosphorylation are molecular on/off switches that regulate a number of key biological pathways within the cell, including signal transduction, cell division, and apoptosis. Phosphatase inhibitors preserve phosphorylation of proteins in cell lysates and tissue specimens. PhosphoGuard™ is specially designed to quench endogenous phosphatase activity in clinical specimens.
The example below shows a xenograft specimen that was split in two equal halves. One half was fixed in regular formalin without PhosphoGuard™ (left panel) and the other half was fixed in PhosphoGuard™ (right panel). The xenograft fixed in PhosphoGuard™ (right panel) shows stronger staining of p-ErbB1 (p-EGFR).
